    Plant-derived compounds as a tool for the control of gastrointestinal nematodes: Modulation of abamectin pharmacological action by carvone

    The combination of synthetic anthelmintics and bioactive phytochemicals may be a pharmacological tool for improving nematode control in livestock. Carvone (R-CNE) has shown in vitro activity against gastrointestinal nematodes; however, the anthelmintic effect of bioactive phytochemicals either alone or combined with synthetic drugs has been little explored in vivo. Here, the pharmacological interaction of abamectin (ABM) and R-CNE was assessed in vitro and in vivo. The efficacy of this combination was evaluated in lambs naturally infected with resistant gastrointestinal nematodes. Additionally, the ligand and molecular docking of both molecules to P-glycoprotein (P-gp) was studied in silico. The presence of R-CNE produced a significant (p 90%. In vitro/in vivo pharmacoparasitological studies are relevant for the knowledge of the interactions and the efficacy of bioactive natural products combined with synthetic anthelmintics. While ADMET (absorption, distribution, metabolism, excretion, and toxicity) predictions and the molecular docking study showed a good interaction between ABM and P-gp, R-CNE does not appear to modulate this efflux protein. Therefore, the pharmacokinetic–pharmacodynamic effect of R-CNE on ABM should be attributed to its effect on membrane permeability. The development of pharmacology-based information is critical for the design of successful strategies for the parasite control.Fil: Miró, María Victoria.     NEMÁTODOS PARÁSITOS DE RHINELLA JIMI (STEVAUX, 2002) (ANURA: BUFONIDAE) EN ÁREAS DE CAATINGA, NORDESTE DE BRASIL

    Los anfibios hospedan una amplia variedad de parásitos, sea de forma temporal (hospedero paraténico), intermediaria o definitiva. Solamente un 8% de los anfibios en Brasil han sido investigados con relación a sus helmintos asociados, por lo que en este trabajo analizamos 20 especímenes de Rhinella jimi Stevaux, 2002 colectados en los estados de Ceará y Rio Grande do Norte, en la región Nordeste de Brasil. Siete taxones de nematodos se encontraron infectando individuos de R. jimi (Oswaldocruzia lopesi Travassos, 1938, Raillietnema spectans Gomes, 1964, Physaloptera sp., Parapharyngodon sp., Rhabdias sp., Cosmocercidae, y una larva no identificada). Todas las especies encontradas representan nuevos registros de parásitos para R. jimi, resultado que amplía los registros de nematodos para esta especie, así como aumentan el conocimiento de la diversidad de parásitos en anuros para América del Sur

    Use of agro-industrial by-products containing tannins for the integrated control of gastrointestinal nematodes in ruminants

    Previous studies have illustrated that different bioactive legume fodders containing condensed tannins might represent one of the options for integrated sustainable control of gastrointestinal nematodes (GIN) in ruminants, which may help address the worldwide development of resistance to synthetic anthelmintics. More recently, impetus has been given to assess the potential antiparasitic activity of less conventional resources, represented by different agro-industrial by-products (AIBPs). This review presents in vitro and in vivo results obtained with a range of tannin-containing AIBPs of various geographical and botanical origins, namely AIBP of nuts, temperate and tropical barks, carob, coffee and cocoa. They tend to confirm the "proof of concept" for their antiparasitic effects and also for other aspects of ruminant production in an agro-ecological context. Socio-economic aspects of the exploitation of such non-conventional resources are also discussed as potential models of the circular economy, by using waste. The different modes of use of these resources are presented in this review, as well as strengths, weaknesses, opportunities, and threats (SWOT) analyses to illustrate the advantages and limitations of on-farm use

    Differences in protein expression associated with ivermectin resistance in Caenorhabditis elegans

    Abstract The indiscriminate administration of synthetic anthelmintics such as ivermectin contributes to the selection of subpopulations capable of resisting the drugs’ effects. To understand the mechanisms of ivermectin resistance in Caenorhabditis elegans, this study attempted to identify molecular targets. C. elegans lineages that were sensitive and resistant to ivermectin were used. Collected nematodes were added to an extraction buffer and macerated in liquid nitrogen for protein extraction. The extracted proteins were separated according to molecular weight by SDS-PAGE to verify their integrity. Subsequently, proteins from both lineages were separated using two-dimensional electrophoresis. The gels were analyzed and the relevant spots were excised and identified by mass spectrometry (NanoESI-Q-TOF and MASCOT®) and subsequently assessed by GO enrichment and STRING® analyses. The increased expression of proteins associated with high metabolic activity, such as ATP-2 and ENOL-1, which are responsible for ATP synthesis, was observed. Furthermore, proteins with involvement in mediating muscular function (MLC-1, ACT-1, and PDI-2), signaling (FAR-1 and FAR-2), and embryo development (VHA-2) were identified.
